酿酒酵母GPDH1基因表达载体pPIC3.5K-ScGPD的构建及其在巴斯德毕赤酵母中的表达
将酿酒酵母3 磷酸甘油脱氢酶基因GPDH1的编码序列构建到表达载体pPIC3.5K中,利用电转化法将构建好的表达载体转入巴斯德毕赤酵母中,利用含有G418的YPD平板筛选到两个阳性转化子,通过分子生物学及外源蛋白表达实验证明,酿酒酵母3 磷酸甘油脱氢酶基因GPDH1已经整合到巴斯德毕赤酵母的基因组上,并且得到了预期的表达.
