miR-412-5p靶向PCDH10对肺癌细胞增殖、迁移、侵袭的影响
目的研究微小RNA-412-5p(miR-412-5p)对原钙黏蛋白10(PCDH10)的靶向关系及其对肺癌细胞增殖、迁移和侵袭的作用。方法实时荧光定量PCR(qPCR)、蛋白质印迹法(Western blot)和免疫组化检测miR-412-5p、PCDH10在肺癌和癌旁组织中的表达。建立抑制miR-412-5p的A549细胞,Western blot检测细胞周期蛋白D1(Cyclin D1)、E-钙黏蛋白(E-cadherin)、基质金属蛋白酶-2(MMP-2)蛋白表达,噻唑蓝(MTT)检测细胞增殖,Transwell检测细胞迁移和侵袭。生物信息学预测和双荧光素酶活性检测分析miR-412-...
